The Effectiveness Investigation of Some Apricot Cultivars on Endoplasmic Reticulum Stress in Different Cancer Cells

Özet

Apricot fruit, which is very rich in terms of high nutritional value and functional food components, is the most produced fruit in the Malatya region and is an important export product. Apricot has a very short shelf life and it is subjected to sulphurisation in order to preserve its current color and increase its shelf life. Despite the advantages of sulphurisation, there are concerns that high sulfur levels may pose health risks. The endoplasmic reticulum is one of the largest of the cytoplasmic organelles in eukaryotic cells. It has significant functions such as calcium storage, detoxification, lipid synthesis and protein folding. Proteins leave this organelle to reach their final destination by folding correctly in the endoplasmic reticulum. Since many diseases, the incidence of which is increasing day by day, such as diabetes, obesity, Alzheimer's, and cancer, endoplasmic reticulum organelle is actually at the center of all these diseases which is thought to be associated with misfolded proteins. Within the scope of this doctoral thesis, the cytotoxic, morphological and cell migration properties of Prunus armeniaca L. Cv. “Hacıhaliloğlu” and “Kabaaşı” apricot species, both as sun-dried and sulphurized methanol extracts, on MCF-7 (human breast cancer) and HCT116 (human colon cancer) cell lines were investigated in vitro. Thapsigargin was used as an endoplasmic reticulum stress inducer. According to the obtained MTT test results, the IC50 values of sun-dried Hacıhaliloğlu, sulphurized Hacıhaliloğlu, sun-dried Kabaaşı and sulphurized Kabaaşı apricots were 60.16, 95.47, 56.32, 108.99 mg/mL for 24 hours against MCF-7 cell line and 79.97, 110.11, 74.94, 117.77 mg/mL for 24 hours against HCT116 cell line, respectively. Fluorescence microscope images of the IC50 apricot concentration values applied in the acridine orange and ethidium bromide staining method and the cell photos taken with the invert microscope as a result of MTT test performed for IC50 determination are completely consistent. According to the results of the cell migration test performed with the wound healing assay, the highest number of cell migration except control group occurred in sun- dried Kabaaşı variety as 9.60%, which was treated with Thapsigargin in the MCF-7 cell line. In HCT116 cell line, the most cell migration occurred as 41.54% in sun-dried Hacıhaliloğlu apricot that was treated with Thapsigargin except control group. When ELISA results for MCF-7 cells are examined, MCF-7 Control group has a higher value on average than all other groups in terms of ATF6 protein. From obtained ELISA results, CHOP protein was not detected in MCF-7 and HCT116 cells. In terms of XBP1 protein, which is involved in another pathway in endoplasmic reticulum stress, HCT116 Control group has a much higher value than all other groups (p<0.05).
