Özet
Amaç: Endometriozis, ektopik endometrial doku yerleşimiyle karakterize kronik bir hastalıktır. Etiyopatogenezi tam
olarak aydınlatılamamıştır. Hastalığın anlaşılabilmesi amacıyla çeşitli deneysel modeller kullanılsa da, cerrahi model
hastalığın anlaşılması ve tedavi etkinliklerini değerlendirmede öncelikli olarak tercih edilmektedir. Bu çalışmanın amacı,
hastalığın mekanizmalarının araştırılmasına olanak tanıyan cerrahi endometriozis modelinin tanımlanmasıdır.
Gereç-Yöntem: Deneysel çalışmalarda düzenli östrus siklusu gösteren erişkin dişi ratlar kullanılır. Anestezi sonrası
abdominal orta hat insizyonu ile karın boşluğuna erişilir. Uterus her iki kornudan çıkarılır ve endometriyal doku, uygun
boyutlarda (yaklaşık 5×5 mm) parçalara ayrılır. Bu dokular steril koşullarda peritoneal yüzeye ya da abdominal duvara
sütür ile tespit edilir. Operasyon sonrası hayvanlar belirli sürelerde (genellikle 2–4 hafta) gözlemlenir ve bu süreçte
endometriozis lezyonlarının oluşumunu desteklemek üzere periton içine düzenli aralıklarla östrojen uygulanır.
Bulgular: Uygulama sonrasında ektopik implantasyon bölgelerinde vaskülarizasyon, lezyon büyüklüğü ve inflamatuvar
yanıt geliştiği gözlemlenir. Histolojik incelemelerde glandüler yapı ve stromal hücrelerin varlığı, modelin başarıyla
oluşturulduğunu gösterir.
Sonuç: Cerrahi model, endometriozisin patofizyolojisini, inflamatuvar yanıtları ve tedavi etkinliğini değerlendirmek için
güvenilir ve translasyonel değeri yüksek bir yöntemdir. Bu model, hastalıkla ilişkili biyobelirteçlerin ve yeni tedavi
stratejilerinin araştırılmasında önemli katkılar sunar.

Objective: Endometriosis is a chronic disease characterized by the ectopic implantation of endometrial tissue. Its
etiopathogenesis has not yet been fully elucidated. Although various experimental models have been employed to better
understand the disease, the surgical model is the most commonly preferred approach for investigating its mechanisms
and evaluating treatment efficacy. The aim of this study is to describe a surgical model of endometriosis that enables
the investigation of disease mechanisms.
Materials-Methods: Adult female rats with regular estrous cycles were used in the experimental procedures. Following
anesthesia, the abdominal cavity was accessed via a midline incision. The uterus was excised from both uterine horns,
and the endometrial tissue was cut into appropriately sized fragments (approximately 5×5 mm). These tissue fragments
were sutured under sterile conditions onto the peritoneal surface or the abdominal wall. After the operation, the animals
were monitored for a defined period (typically 2–4 weeks), during which estrogen was administered intraperitoneally at
regular intervals to promote the development of endometriotic lesions.
Results: Following the procedure, vascularization, lesion enlargement, and inflammatory responses were observed at44the sites of ectopic implantation. Histological examinations revealed the presence of glandular structures and stromal
cells, confirming the successful establishment of the model.
Conclusion: The surgical model is a reliable and translationally relevant method for studying the pathophysiology of
endometriosis, associated inflammatory responses, and the effectiveness of therapeutic interventions. This model offers
significant advantages for investigating disease-related biomarkers and developing novel treatment strategies.
