Investigation of the Effect of Exercise on Nuclear Factor-ΚB Ligand, Osteoprotegerin and Irisin Levels in an Experimental Hypertension Model

Özet

AIM: The nuclear factor-ΚB ligand receptor activator (RANKL), nuclear factor-ΚB receptor (RANK), and soluble decoy receptor osteoprotegerin (OPG) (RANK/RANKL/OPG) pathways, as well as the myokine irisin, have been reported to play a role in the pathophysiology of cardiovascular diseases. Our study aimed to investigate the roles of RANK, RANKL, OPG, and irisin in the pathophysiology of hypertension and the effects of exercise on RANK, RANKL, OPG, and irisin levels.

METHODS: In the study, Sprague Dawley male rats were divided into control, control+exercise, hypertension and hypertension+exercise (n=8/group) groups. Hypertension and hypertension+exercise groups were given L-NAME (400 mg/L) with drinking water for 6 weeks. Control+exercise, hypertension+exercise groups were made to exercise at a speed of 20 m/min for 45 min for the last 4 weeks. After 24-hour urine collection; blood, muscle, heart and kidney tissues were taken under xylazine (10 mg/kg) and ketamine (50 mg/kg) anesthesia and euthanasia was performed. Data were analyzed with Student's t-test and Mann-Whitney U (p<0.05).

RESULTS: Compared to the control group; GSH levels decreased in the muscles and increased in the heart in the control+exercise group (p<0.05), mean arterial blood pressure, heart and kidney GSH levels and serum irisin level increased in the hypertension group (p<0.05). Compared to the hypertension group; MDA muscle level increased in the hypertension+exercise group (p<0.05).

CONCLUSIONS: Our findings suggest that exercise may be effective in the physiopathology of hypertension through irisin levels and oxidative stress parameters. It is thought that the effects of the RANK/RANKL/OPG pathway on hypertension require further studies with different experimental and clinical studies.
